ISRIB (Integrated Stress Response Inhibitor)

ISRIB (Integrated Stress Response Inhibitor) is a synthetic small-molecule compound studied for its potent ability to modulate cellular stress signaling by counteracting the activation of the integrated stress response (ISR). It is widely utilized in laboratory research focused on translational regulation, protein synthesis, and cellular resilience mechanisms under various stress conditions.

Technical Specifications

Mechanism of Action & Research Utility

ISRIB functions by enhancing the activity of eukaryotic initiation factor 2B (eIF2B), the guanine nucleotide exchange factor essential for translation initiation. During cellular stress, the phosphorylation of eIF2α typically suppresses eIF2B activity, thereby reducing global protein synthesis. ISRIB restores translational activity by stabilizing eIF2B in an active conformation, even in the presence of eIF2α phosphorylation.

Primary fields of in vitro laboratory investigation include:

• Integrated Stress Response (ISR) Signaling: Probing the pathways governing cellular responses to stress.

• Translational Control: Mapping mechanisms of protein synthesis regulation.

• Proteostasis: Studying cellular adaptation to metabolic and proteotoxic stress.

• Cognitive Research: Investigating synaptic plasticity and memory pathways in neurobiological models.
