Flmodafinil (CRL-40,940)

Flmodafinil, documented in neuropharmacology literature as CRL-40,940 (and colloquially termed Lauflumide), is a synthetic, bis-fluorinated eugeroic compound structurally related to modafinil. It is characterized by the integration of twin para-fluorine atom substitutions positioned on its core benzhydryl ring matrix, linked directly to a terminal sulfinylacetamide chain. Within translational neuroscience and behavioral modeling, flmodafinil serves as a high-affinity chemical tool for investigating the mechanics of central alertness, sleep-wake architecture, and targeted monoaminergic reuptake inhibition. The strategic halocarbon modifications alter the compound’s electron density distribution, which researchers study for its impact on metabolic persistence and central nervous system (CNS) partition coefficients compared to unhalogenated parent molecules.

Structural Attributes to Note: The analytical diagram above highlights the specific bis(4-fluorophenyl)methylsulfinyl architecture that defines Flmodafinil. The central sulfoxide center (-S=O) acts as a chiral axis, flanked by a primary acetamide segment and a di-fluorinated benzhydryl backbone. The robust carbon-fluorine bonds are heavily evaluated in biochemical stability assays to observe how electronic shifts affect resistance against traditional hepatic amide-cleavage and oxidative degradation pathways.

Research Applications & Mechanism of Interest

The utility of flmodafinil within modern behavioral models and cell-free matrices focuses on its capacity to safely upregulate extracellular signaling vectors without inducing peripheral sympathomimetic cascades.

Primary fields of in vitro and preclinical laboratory investigation include:

• Dopamine Transporter (DAT) Affinity Kinetics: Measuring competitive binding assays at the dopamine transporter site. Investigators use flmodafinil to study the slowing of monoamine reuptake, resulting in prolonged synaptic presence of endogenous dopamine.

• Vigilance & Locomotor Baseline Architecture: Utilizing real-time polysomnographic and actigraphy tracking in rodent cohorts to chart modifications in deep non-REM/REM sleep intervals and baseline target attention metrics.

• Orexinergic Coordination Interception: Investigating downstream cross-regulation pathways within the lateral hypothalamus to observe how DAT-mediated signaling correlates with the release profiles of orexin-A and orexin-B peptides.

• Comparative Isomeric Structure-Activity Relationships (SAR): Serving as a vital comparative standard alongside modafinil, armodafinil, and adrafinil to map how localized halogen substitution patterns steer receptor selectivity profiles.

Storage & Handling Guidelines

Store at controlled room temperature. Keep tightly closed. Liquid formats: do not refrigerate or freeze. Cold storage may cause the solution to cloud or precipitate. If this occurs, return to room temperature and mix until clear.

Storage guidance is a house recommendation. Analytical documentation is per-lot release testing.
